pMEV2HA-cRAF1-wt
Human cRAF Xpress Clones express cRAF (Raf-1), a serine/threonine kinase involved in RAS signaling pathways. It functions downstream of RAS GTPases and activates MEK1/MEK2, which in turn phosphorylate and activate ERK1/ERK2. These activated ERKs regulate cell division, apoptosis, differentiation, and migration. The c-terminal kinase domain of cRAF (residues 305-648) is cloned into the pMEV-2HA vector. The K375M mutation, which replaces the essential lysine 375 with methionine, inactivates the kinase, creating a dominant-negative inhibitor that can block the function of endogenous cRAF in mammalian cells.
