Human NIK Xpress Clones express NIK (NF-kB-inducing kinase), a serine/threonine kinase involved in activating the NF-kB pathway, a key mediator of inflammation and immune responses. NIK, a 947-amino acid protein, binds to TRAF2 to stimulate NF-kB activity and is part of a signaling cascade common to TNF/NGF receptors and IL-1 type-I receptors. The kinase's K429 is essential for activity, and the K429/430A mutant is kinase-deficient, serving as a dominant-negative inhibitor of NIK signaling. Additionally, phosphorylation of T559 activates NIK, while the T559A mutation prevents its activation and IKKα phosphorylation, interfering with TNF-α-induced NF-kB activation.
